Remove impediment. Remove tire valve core (tool provided with the slime bottle). Squirt in required amount of slime. Clean inside of valve stem. Re-insert valve core. Inflate to regular pressure. Either go for a drive right away, or put it on a rear stand and run it in 6th gear at idle. Do NOT let it sit for a period of time between squirting the slime in and bringing it up to pressure and rotating it.
You might notice for the first few mins of your ride the tire feels unbalanced.
80ml for your front tire or 157ml for the rear.

Tire pressure is same as before and last I rode was 12-14 hrs ago, so I don't think it's deflating at all.
Should I leave it there until new tire arrives ?
Or should I pull it out and see if it really punched a hole ?

He puts some really good patches. no need to buy a new tire.
If it was me, i would remove the screw, put a plug on it (purchased at CT or any other store) and ride it to Dave
get some soapy water, and apply it on and around the object and move it around with a little wiggling. Look for bubbles, to determine if it has actually punctured...I do this before anything to confrim its actually punctured...Sometimes you get lucky and the object is not long enough to puncture, or goes in sideways and doesnt puncture.
Your tire looks so new, I would not buy a new one...I would install a plug/patch from the inside of the tire....They are cheap and last long with no chance of falling out like a regular rope plug that is only temporary

Tire slime is good in a real emergency, like if you're stranded 600miles between Guadalupe Death Trail and The Brownair Desert, and your tire plug kit fails first.

1. The nut came most likely from my street due to construction here. So, I may get it again until the construction is over.
2. I'd shoot myself if I got another screw in the new tire again.
