Without even seeing the bike ...
There are only two hoses on the fuel tap ... one is the fuel line, the other is the vacuum port. They're different sizes. You can't mix them up.
There is a vacuum port on one, or both, carburetors. It will be on top adjacent to the cylinder head. If only one carburetor has this connection, that's where the hose from the fuel tap goes. If both have it and one of them has a rubber cap over it, the hose goes to the other one. It makes no difference whether it's the left or the right cylinder, it will work the same, so connect to whichever one that the hose reaches. If you need to make up a new hose ... it makes no difference which cylinder you connect it to. Just make sure the other vacuum port is capped.
There is a remote possibility that both carburetors have a hose connected to that fitting, which leads to a T-fitting, which in turn leads to the fuel tap.
